The definitive start of active combat occurs when Kabuto activates his and summons an army of legendary deceased shinobi (including former Kage, Jinchuriki, and Akatsuki members). Simultaneously, Tobi unleashes the Gedo Statue and the 100,000 White Zetsu from the Demonic Statue of the Outer Path.
